The interaction effects of intra-cerebral injection of the crocin with the α2-adrenoceptors antagonist on anxiety behaviors in chronic pain in rat

Introductions: Neuropathic pain is one of the most important types of chronic pain and in these patients, anxiety is one of the common co-morbidities. In traditional medicine, saffron is used for its beneficial therapeutic effects in relieving pain and anxiety. This study aimed to investigate the effects of crocin injection (saffron active ingredient) on anxiety, following induction of neuropathic pain caused by chronic constriction injury (CCI) of the sciatic nerve in rats. The α2-adrenoceptors also have an important role in modulating pain and anxiety. How the new analgesic herbal compound mediates pain behaviors via interaction with α2-adrenoceptors remains unclear. In the present study, we also investigated the interaction effects of crocin with central α2-adrenoceptors on anxiety following CCI.
Methods and Materials: After cannulation inside the lateral ventricle and a three-day recovery period, on the 4th day, the neuropathic surgical model was created. Drugs injection started from the day of neuropathic surgery and continued for fifteen days (Crocin 40μg/μl, α2- antagonist Yohimbin 10μg/μl). Anxiety behaviors were assessed by using the EPM and open field test on days -5, 2, 4, 6, and 10. The statistical analysis of data was performed by analysis of variance (one-way & two-way ANOVA) followed by Tukey post-hoc analysis. In all cases, differences were considered significant if p<0.05.
Results: The findings of this study showed that CCI caused anxiety behaviors. Crocin has anxiolytic effects. Co-injection of yohimbine with crocin attenuated the crocin anxiolytic effects.
Conclusion: Our study provided information about the anxiolytic effects of crocin. Additionally, it seems that the anxiolytic effects of crocin are mediated by α2- adrenoceptor in rats with chronic pain.
