Downregulation of proenkephalin gene in the hippocampus of depressed male rats and its modulation during treatment with niosome hesperidin

Introduction: Depression is a debilitating mental disorder related to cognitive impairment. In this study, we aim to investigate the downregulation of the proenkephalin gene in the hippocampus of depressed male rats and its adjustment during treatment with noisome-hesperidin.
Methods and Materials: This study experimented on 36 adult male rats, which we divided into six groups. Control group, depressed group, hesperidin group, depressed group which was treated by hesperidin, niosome-hesperidin group, and depressed group treated by niosome-hesperidin. Then we used the PCR method to evaluate the proenkephalin gene level in the hippocampus of different groups.
Results: The results of this study show that the level of proenkephalin gene in the control group and the depressed group has significantly decreased compared to the depressed+hesperidin group (P<0.001). Also, the level of this gene in the depressed group+hesperidin increased significantly compared to the depressed group+niosome-hesperidin, the hesperidin group, and the niosome-hesperidin group (P<0.001).
Conclusion: The results of this study show that depression causes a decrease in proenkephalin gene level; while in the group treated with hesperidine, the level of this gene increased. This confirms the decrease in proenkephalin gene expression during the depression and the increase in its expression during treatment with niosome-hesperidin.
