Introduction: Mitochondria play a central role in energy metabolism and signaling pathways. In this study, the effect of MitoQ supplementation alone and in combination with endurance training (ET) or high-intensity interval training (HIIT) was investigated on skeletal and cardiac muscle of male rats on the process of mitochondrial quality control (fission and fusion). Methods and Materials: The animals were assigned into six groups (n=7): control, MitoQ, ET, ET+MitoQ, HIIT, and HIIT+MitoQ. In training groups, rats were trained on a treadmill for 8 weeks, 5 days/week. MitoQ was administered in drinking water. The gene and protein expression were measured by Real-Time PCR (2-ΔΔCT) and western blot analysis, respectively. Results: In skeletal muscle, performing ET significantly increased Drp1 (P<0.0001) and Mfn1 (P=0.0003) proteins and Opa1 gene expression compared with the control group (P=0.02). HIIT only increased Mfn1 protein levels compared with the control group (P<0.0001). MitoQ, in combination with HIIT, significantly increased Drp1 expression in skeletal muscle compared to the MitoQ alone. Conclusion: In skeletal muscle, ET was more effective than HIIT. HIIT by regulating proteins of mitochondrial dynamics, improves the metabolic states and heart energy. In the heart tissue, the combination of MitoQ with ET has more effective results than MitoQW+HIIT, and no derange in mitochondrial dynamics.
