Introduction: Alzheimer's disease is a common neurodegenerative disease that causes cognitive impairment and memory loss. Chronic neuroinflammation and the loss of cholinergic pathways are two of the main pathogenic factors leading to neuronal damage. Donepezil, an acetylcholinesterase inhibitor, is effective in improving the symptoms of Alzheimer’s disease, but has a low tendency towards butyrylcholinesterase; therefore, new therapeutic strategies with increased response rates are needed. Trans-cinnamaldehyde (TCA), a bioactive compound of cinnamon, has been shown to have anti-inflammatory and neuroprotective effects in various studies. In this study, we aimed to investigate the potential synergistic effects of TCA and donepezil on memory improvement in mice with scopolamine-induced memory impairment. Methods and Materials: 48 adult male mice were divided into eight groups: the first group received normal saline, the second group TCA 40 mg/kg, the third group TCA 60 mg/kg, the fourth group Donepezil 1 mg/kg, the fifth group Donepezil 3 mg/kg, the sixth group TCA 40 mg/kg and Donepezil 1 mg/kg, the seventh group TCA 60 mg/kg and Donepezil 1 mg/kg, and the eighth group received TCA 60 mg/kg and Donepezil 3 mg/kg via intraperitoneal injection. The mice's memory was impaired with scopolamine 1 mg/kg. After the treatment period, their spatial memory was assessed using the shuttle box apparatus. The data was analyzed using one-way ANOVA with SPSS 21. Results: The combination of donepezil 1 mg/kg with TCA 40 mg/kg and TCA 60 mg/kg had a significant effect on improving the memory of mice compared to the control group (P<0.001). Conclusion: In conclusion, our study suggests that the combination of TCA and donepezil have synergistic effects and could be viewed as a promising therapeutic approach for the treatment of Alzheimer’s disease.
