The effect of conditioned medium obtained from undifferentiated mesenchymal stem cells derived from adipose tissue on the heart injury caused by Movento in male rats

Introduction: Spirotetramat is a keto-enol compound that is used as an insecticide for sucking pests in various crops such as pistachios, citrus fruits, tomatoes, etc. Movento contains 400 g/L of spirotetramat, which can cause severe poisoning in humans and rats through oral consumption. The aim of this study was to investigate the effects of conditioned medium obtained from undifferentiated mesenchymal stem cells (CM-SMC) on the histological and functional disorders of the heart due to the oral consumption of Monto in male rats.
Methods and Materials: In this study, 30 male rats (n=6) were weighed and randomly divided into 5 groups: Control (CTL), Movento (MOV), MOV+Payeh (P), MOV+codition media (CM), MOV+CM+P. In the Movento groups, the animals were gavaged with 1000 mg/kg of Movento (1 cc in volume) for 7 subsequent days (once every 2 days). The culture medium obtained from human fat tissue after optimization and enrichment and also payeh, will be injected through the tail vein of the animal (15 microliters). On the 10th day, the animals were anesthetized and the mean arterial pressure was recorded with a Powerlab device. Then the heart was removed for pathological work.
Results: The mean arterial pressure in the rats receiving Movento was significantly reduced compared to the control group (p<0.05). Enriched culture medium restored this reduction (p<0.05). Also, the inflammation increased in the heart tissue of the Movento group (score 3) compared to the control group (p<0.01), and reduced to lower scores by the culture medium treatment (p<0.01). Payeh had no effect on pressure and inflammation (p>0.05).
Conclusion: CM-SMC ameliorated the inflammation and mean arterial pressure in male rats. Therefore, it seems that the enriched CM can have a good effectiveness in solving heart problems caused by Movento.
