اثر تمرینات منظم شنا بر یادگیری و حافظه فضایی و مصرف اختیاری مورفین در بچه موشهای جداشده از مادر در بزرگسالی

هدف: جدایی از مادر میتواند موجب اختلال در یادگیری و حافظه و افزایش احتمال ابتلا به اعتیاد شود. از سوی دیگر، تمرینات فیزیکی ممکن است موجب تغییرات بلندمدت در مسیر پاداش و هیپوکمپ گردد. لذا مطالعهی حاضر با هدف بررسی اثر ورزش شنا بر یادگیری و حافظه فضایی و مصرف اختیاری مورفین در رتهای نر و ماده جداشده از مادر انجام شد.
مواد و روشها: بچه موشها در طی روزهای 2 تا 15 پس از تولد، هر روز به مدت سه ساعت از مادرشان جدا شدند و نیز در روز 21 از شیر گرفته شدند. رتهای گروههای ورزشی روزانه یک ساعت و در روزهای 57-28 پس از تولد در معرض ورزش شنا قرار گرفتند. در پایان این دوره، برای سنجش یادگیری و حافظه فضایی از ماز آبی موریس، و برای ارزیابی میزان مصرف اختیاری مورفین از مدل انتخاب دو بطری استفاده شد.
یافتهها: موشهای جداشده از مادر در هر دو جنس، نسبت به موشهای گروه کنترل، زمان طولانیتری در رسیدن به سکو در فاز یادگیری نشان دادند. ورزش شنا در موشهای جداشده از مادر، موجب کوتاه شدن زمان رسیدن به سکو گردید. جداسازی از مادر و ورزش شنا تاثیری بر حافظه فضایی در تست حافظه نداشت. همچنین، میزان مصرف اختیاری مورفین در بچه موشهای نر و ماده جداشده از مادر نسبت به گروه کنترل بالاتر بود، که ورزش شنا موجب کاهش آن گردید.
نتیجهگیری: شنا ممکن است برای پیشگیری از سوءمصرف مواد و بازتوانی اختلالات شناختی به دنبال جدایی از مادر سودمند باشد.
Introduction: Maternal separation (MS) can lead to learning and memory deficits, and also the risk of addiction. On the other hand, physical activities may lead to long-term changes in the mesolimbic dopamine pathway and the hippocampus. This study was designed to examine the effect of swimming exercise on the spatial learning and memory and voluntary morphine consumption in maternally separated male and female rats.
Materials and Methods: Rat pups were separated daily from their mothers for 3h during postnatal days 2–15. All pups were weaned on day 21. The exercising rats were exposed to swimming exercise for 1 hour per day during postnatal days 28–57. At the end of this period, rats were tested for the spatial learning and memory in the Morris Water Maze, and voluntary morphine consumption using a two‐bottle choice paradigm.
Results: We found that the MS male and female pups showed longer escape latencies to find the platform as training progressed than No MS/no swim rats. Also, swimming exercise shortened the latency to escape in the male and female MS pups than MS/no swim pups. MS and swimming had no effect on spatial memory in the probe test. Also, voluntary morphine consumption was higher in the male and female MS/no swim than No MS/no swim pups, while swimming exercise decreased it.
Conclusion: Our results have shown that swimming exercise during adolescence may be of benefit for prevention of drug abuse and recovery from cognitive deficits following neonatal MS.
