Activity of biofilm genes icaA and icaR in methicillin-resistant Staphylococcus aureus treated with vitamin K in wound specimens

Introduction: The use of vitamin K plays a very important role in human health. This vitamin may have effects on some of the pathogens such as biofilms in Staphylococcus aureus.In this account, the purpose of this study was to determine the effect of vitamin K on the expression of icaA and icaR genes in methicillin-resistant S. aureus isolated from the wound sample.
Materials and Methods: In this interventional study, 15 isolates of methicillin-resistant S. aureus were treated with vitamin K at concentrations of 1, 10, 50, 100, 200, 300 and 500 μg / ml. Time intervals for incubation were determined  24, 48 and 72 hours. Finally, the desired RNA was extracted and cDNA was synthesized. The expressions of icaA and icaR genes were evaluated using qPCR method.
Results: The expression levels of icaA and icaR genes were used as controls prior to vitamin treatment. Both of genes showed a reduced expression rate in isolates which treated by vitamin K. This effect on the expression of icaA and icaR was clearly observed at a concentration of 200 μg and also in the 48-hour incubation.
Conclusion: Vitamin K along with some antibiotics could be effective against some certain skin infections which caused by methicillin resistance S. aureus.
